The short answer is: it doesn't work that way

The long answer: you need the original setup.exe and all the ancillary files (i.e. the original CD). You may as well find it.

It is possible that you might be able to copy the program the way that you want but you will have to be very careful to get all the files that it needs to run.

First off open up any folder and go into Tools -> Folder Options. Click the 'View' tab and look down the list until you find the 'Hidden Files and Folders' option - change it to show hidden files and folder.

Now go into My Computer -> C Drive (or whatever your main harddrive is) -> Program Files. In here you should find a folder for the program you're trying to transfer. Start by copying that folder onto the memory stick.

Now look in My Documents and see of there's a folder in there that's related to your program - if there is copy that as well.

Finally go into C:\Documents and Settings, find your user file (will have your name or Administrator on it) and then go into Application Data and see if there's a folder in there for your program and copy it if there is.

Now plug the memory stick into the second computer and put all those files back where they came from. If you're very lucky the program will run. If not then the installer put some files in shared places or changed environment variables and that's not something that you can fix easily so you may be out of luck.

You have to transfer the program by copying the program installer into the memory stick, not just the shortcut icon. After you copy it to another computer, you have to reinstall the program in that computer to make it work. ^_^
