If you right click on a folder goto properties > security you are shown a list of everyone who can access that folder. To just allow certain people access to that folder remove the everyone account from the list and any people you do not want to allow into the folder. Always leave the system in there and the administrator and creator/owner accounts. Then add only the people you wish to allow access to the folder into the list. Make sure you give each person the proper rights when accessing the folder or they may not be able to list, read, or execute files within it.

Yes you can create a hidden share and also secure it.

While creating a shared folder, simply add $ sign at end of the share name, moreover, you can also goto the -folder properties -> security tab and setup up access level permission each different users and groups in the domain.
