It's both a play and a player, oddly enough. The play is when an offensive player is tackled all the way back in his own end zone. Don't forget, the offense is trying to score by getting into the OTHER team's end-zone; being in your own end zone is a bad situation. There are a couple of other plays that can result in a safety, but that's the basic one - the offense is pinned so far back on the field that the defense is able to tackle the ball carrier in his own end zone. The defense thus earns 2-points for its team.

As to the player position known as safety, it is a defensive player who generally plays pretty far back off the line of scrimmage. Generally, for most plays, a team will line up 2 safeties, and their job is to make sure nobody gets behind them. They are the defense's last resort; the last stand. Thus they are called "safeties" as a conservative defensive measure just in case the offensive player gets past the first two lines of defense.

The safety plays defensive back. He is kind of like the last line of defense. He usually starts about 10-15 yards off the line of scrimmage and will either drop back into a deep zone or guard a wide receiver that is in the slot.

It depends do you mean the penalty because that means some one is tackled in there own endzone and the other team gets to points and the ball. The positions a the person furthest back and doesn't let anyone get behind them and is an overall great defensive player.
