Tricky question. You got some answers that refer to Alexander the Great and Genghis Kahn, which refer to empires rather than countries with defined borders. In their times, Macedonia and Mongolia were defined as regions and not countries; if those count, then definitely the largest would have been England in around the mid-18th century, which had an empire that included Canada, Australia, India, eastern Australia. I would disqualify all of these.
The USSR would have been the largest country, or if you want to argue that the USSR was a union of individual countries (as the Soviets claimed), then Russia was and still is the largest country that ever existed.
