I would put it in a folder. Go to Office Max and find a really nice folder, don't get a bright or childish folder. You just need something nice and simple and professional-looking.

I'll probably go against the flow here.
Put it in a nice leather portfolio that you would keep your interview notes in and just hand them the paper at the start of the interview, if they don't already have a printed copy. Nothing messes up my files more than having the little covers sticking out, so I wind up tossing the covers. In fact, I cannot remember being impressed by a cover, and to my knowledge I only ever hired ONE person who put their resume in a cover.
Make sure the paper is a nice parchment or linen, and your formatting impeccable!
The only time I would say to put a cover on is if you are applying for a PR or Marketing or Design position. Then appearance is everything.

If you are looking at a job in an office environment, professionalism is a must. Put it in a clear soft folder, it will make it stand out (won't get lost in with all the others) and they are very cost effective. It may also pay to have your photo copied on to it so that they remember who you are. Just make sure you mention you experience, previous jobs, how great you are, schooling and references. Never more than 2 pieces of paper. Also, don't include any jobs that are not relevant to the position.
Good luck, hope you get a great job.

There are resume covers that you can buy at Office Depot. Just make sure that the resume page matchs the color of the cover. It's not necessary to put it in a cover though.
