I am assuming you are looking for an easy way to do this without permission settings and all that you would have in a corporate environment. Try google search on Microsoft Private Folder 1.0. Its a very small utlity made by microsoft itself. It is very easy to use and will lock a folder with a password.

Set the folder security to only be accessible by a certain account or group. I'm not aware of a way to set a password on a folder.
