If you are running Windows XP, or not too much older , go to Start > All Programs > Accessories > System Tools > click on " Disc Defragmenter". This will take a little while if you have tons of stuff on your C-drive, but, it will gather all the fragmented parts of programs & files & put them in one place, so when you want to bring up a program, it takes a lot less time for the computer to find all the parts. I just did that, after I used the DISC CLEANER, found in the same place as the defragmenter. The cleaner will compress little used files & programs, gaining you more space on your drive.

If you are running Windows XP, go to Start>All Programs>Accessories>System Tools>Disk Defragmenter and follow instructions.
Kiya is also correct in rebooting into Safe Mode.
Safe Mode only runs the minimal drivers so the defrag is completed quicker.
The F5 works as a key to get into safe mode boot menu as well.
Make sure you delete your internet cache weekly.

ok, id like to say that defraging your computer will only increase your computers 'finding' speed. when you have all sorts of files scatered around your computer defraging it puts them back in there place.defraging takes way too long and i have a better way
more files and how big the files are arent the main cuase of you computer bieng slow.
the main cuase is 'running processes'
its all the stuff thats running NOW
theres a question that i asked on this, heres a link to it:
http://answers.yahoo.com/question/index;_ylt=AiouRC9qV03_s_aEESQytV3sy6IX?qid=20070218042153AAyvv0t
u need to stop these processes frm running at start up
i cant be bothered to type anymore so type in google 'how to stop programs frm running at startup'
or ask yahoo answers
trust me defraging doesnt make much of a diff
hope this helps
:)

this is how i do it. it may be easier for you.
click your My Computer icon,
then Control Panel,
then Administrative Tools,
then Computer Management,
there you'll find disk defragment, just follow the instructions.
good luck! by the way, you should do this at least once a month.

I called tech support to talk me through reinstalling my drives and now it works as well as when it was first bought. ALSO - some things slow down your computer like INSTANT MESSENGER and PHOTOS AND SCREENSAVERS. Try getting rid of that stuff if you have been using them, and see how it runs. Also, click on 'Tools' at the top and go to 'internet options' and click on 'delete cookies.' That helps, too.

Open it in Safe Mode....run your defrag...shutdown and return to Normal. To run in SF turn your comp. off to REBOOT..not shutdown...and when it starts to reboots keep hitting your F8 key to get to selections to run.
