I'd say many thousands, but it'd be nearly impossible to quantify. There are general regions that encompass several states each like the Pacific northwest (West), the southwest, New England (Northeast), Plains/midwest (all of the middle),and the South (southeast). There are also 50 states, with each state having many regions.
For example: Belmont, California is in central region of San Mateo County. San Mateo County is in what is known as the peninsula region of the San Francisco Bay Area. The SF Bay Area is in Northern California, which is in turn a region of California. California is often considered part of either part of the Pacific Northwest, the Southwest, or just the Pacific Coast.
Many cities even have regions in them. San Francisco has many neighborhoods (Pacific Heights, Mission, Nob Hill, etc.) Even these neighborhoods have different regions (Lower Pacific Heights, outer Mission, lower Nob Hill, etc.)
So really, unless you're more specific, the answer is anything from a few to thousands.

The Census Bureau defines 4 major regions: West, Midwest, South, and Northeast. Those are divided into a total of 9 subregions.
But 'region' is a very relative term, and different people will tell you wildly different answers.

Regions is a variable because there are many definitions,
What are you really asking ?
Mountain vs plains ? Desert vs wet ? Urban vs country ?
Economic regions like manufacturing or farming ?
There are hundreds of regions depending on your definition.

You can make about as many as you want...
By terrain, location, waterways, proximity to other countries (i.e. Canada and Mexico).
I am from Minnesota. Most people call it the midwest or the upper midwest. Some call it the great plains, the Northern plains, Great Lakes area... it goes on and on.
