First you would create separate user accounts and make sure the other users is not an admin.
The right click on the folder and from the security tab apply the permissions/restrictions.
When that user tries accessing that folder they will get an error.

u hav to use a softwares which lock folders using the kernel (this means u cannot use them in vista, since vista's kernel is locked) use softwares like folderguard, folderlock, folderpassword etc.
