Polyester is man-made (synthetic) comprised of a variety of different materials. Cotton is natural, and grown in fields.

Cotton is a natural fabric commonly used to make t-shirts, jeans, socks and underwear. It is extremely absorbent and strong.
Polyester is an artificial fabric (made from many [poly] esters). Probably the most common use for it is blended with cotton in a t-shirt... polyester/cotton blend is a cheaper fabric than cotton and it shrinks less than cotton alone. Basketball shorts are almost always made out of pure polyester.
