In a very simplified manner, the defense trys to stop the opposing teams offense from scoring. They do this by trying to allow the offense to advance the ball as little as possible, while at the same time trying to steal the ball from them. If they tackle the quaterback, thus making the opposing team loose yards then all the better.

It depends on which team your talking about. If you were referring to the Detroit Lions...what their defense does is take the opposing offense by the hand and gingerly walk them 80+ yards downfield, then they make sure that the guy with the ball has at least a six foot invisible force field around him so that there is absolutely no chance that he will not score. This is not the original way defense was designed to operate.
