Yes, but I would be more concerned about Vista support for the actual device you plan to attach to this port.

Yes, all version of Windows Vista support RS232. RS232 is the Serial Port Standard. But be careful because a lot of new computers and especially laptops are not shipping with serial ports anymore.

They are going in favour of USB and abandoning the PS/2 and Serial ports.
