As far as I know, the mission specialist is usually the person who designed/built whatever they are launching/using. I have a friend who was a mission specialist/astronaut - they were launching the satellite he was the Principal Investigator on. It's a lot of hard work, Ph.D. in physics or astrophysics and you have to be able and willing to lead a group and spend YEARS on one project. Very few people with the qualifications will ever get the opportunity.

Have you had a look around NASA's website? Or just type 'astronaut qualifications' into a search engine and get several thousand hits to go wade through.

2. Be a super duper pilot and get chosen for the space programme.
3. Win the lottery and pay to go.
