There are two ways you can do this:
One: Before you leave your desk, hit control+ALT+delete all at the same time. When you come back to your desk, hit control+ALT+delete, and enter your password.
Two: If you don't want to have to remember to lock your desktop every single time you walk away, you can have your desktop lock itself after a certain amount of time.
You can do this by going to your control panet. Then go to Display. Then click on Screensaver. You can choose how many minutes you would like for your screensaver to stay as it is before it locks itself. Make sure you click in the check box to "resume password protect". This makes it so only you can unlock the computer with your password. Then click ok.
I like this method better because often I have to leave my desk at all times of the day, and it's easy to forget to lock my desktop. This way I don't have to think about it at all!

To do this hit Ctrl Alt + Delete as if you were logging on then press the button marked Lock Computer. To unlock the computer you just hit Ctrl Alt + Delete again then put in your password.

Save anything first, then CTRL, ALT and deleat at the same time. this will go to an option screen then click lock worktop, workstaion etc. etc. they come in many shapes and forms. Also always save before locking as turning off the pc simple unlocks it.

Depends on the Operating System.
Windows NT based: Ctrl Alt Del - Lock
Windows XP based: Log off + Accounts Password protected
Windows XP 2: Win Key + L
That's all I know of.

First you will want to go to the START button
once you have pressed that
at the very bottom of the start menu you will see
2 buttons
Log Off & Turn Off Computer
From hear you have 2 opiotions
you can ether just press log off (Thu if you have a password you will have to enter it every time)
Or you can press Turn Off Computer
From there 3 choices will appear
Stand By & Turn Off & Restart
Press Stand By
This will Temperarely turn off your computer all you should have to do to wake it up is move the mouse from side to side of press the space bar on your keyboard
thats it

well there is the ugly way and the novice way...
will start from the ugly way... you'll need to chage a registry value which will enable the screen which popups when you click Ctrl +alt+delete appear and you then you can lock it... as for which registry value... well i can't remember.. but i do know it works cos i did it...
nice wy... well use a windows tweak software to enable that option...

right click on desktop > properties > screensaver tab > "on resume password protect" should be enabled and you can set a password on your account from start > control panel> user accounts> go to your account and you will see password. or instantly you can do cntrl alt delete
