Actually I watched part of the combine yesterday and Deion basically confirmed he ran a 4.19.
I'm unsure if that is the fastest ever, but I'd say it's a safe assumption. Ofcourse that was by hand held stop watch.
To the best of my knowledge the fastest electronically recorded time is by Rondel Menendez WR, Eastern Kentucky in 1999, he ran a 4.24.
Although, it's claimed that electronically timed 40's are actually slower than the times really are. Part of this is because it goes on any motion, like moving your swing arm a little early.

Deion had the fastest 40, probably. but there are technicalities on the way in which certain times where timed(electronic or manual) so theres disputes going around about whos fastest,fabian washington was the fastest ever in the combine at 4.25(electronically) but the reaction times involved in hand timing mean deion was prob faster at 4.29 manual

Actually, no one knows for sure. Electronic timing (and the NFL Network) weren't around until last several years. So, you'd have to take a player's word for it.
Deion didn't confirm his 4.19 or 4.20 time on the NFL Network, but he didn't deny it either. I wrote 2 stories about this on my blog. Check them out.

I was under the impression it was CB Fabian Washington 2 years ago w/a 4.25
and this year it was WR Yamon Figurs w/4.30 (^Adrian Peterson ran a 4.40)
