Basically the President then nominates a successor that has to be approved by Congress. You can find details of this in the 25th Amendment to the Constitution.

The president would appoint a new Vice President, whom the legislative branch would ratify. In the meantime, if something happened to the President, I believe the speaker of the House (currently Nancy Pelosi) would be the next person in line.
