To state that marketing is manipulative is like saying the sun is a star. Of course it is. To be manipulative is to change something to fit your will. To market something is to tailor a product and product delivery method that caters to the customers will and tries to create as many new customers as possible. To create a new customer you must convince some one to establish a new behavior. This behavior is different from previous behavior. If it is inspired by a commercial or an ad campaign, then the person has, by definition, been manipulated. The question seems to fall in one of three categories, is it ethical (the one I think you might be leaning towards), why is it effective or how does it work. Any one of these will provide a starting point for a great paper. Just remember, just because some one is being manipulated does not mean they are taken advantage of. If you are going to address the ethical issues of marketing, pick out what areas are unethical and why it is.
Just saying it's manipulative and trying to let the pejorative undertones of the phrase make the argument for you is not enough for an intelligent discourse.

to answer the last part of your question first I believe that to manipulate a thing you must first understand a thing so there is a bit of both they are understanding why you buy so they can make you buy their products/ services. As for the first part I would go with How marketers manipulate people and why it works.

I think he is concerned that your paper will sound like more of an opinion piece than a research paper. I agree they are trying to manipulate, but your paper will sound better if you don't say it that way.

Absolutely... I think you've found a very interesting subject... How will you gather the information? I suggest experimentation... The results could be very very interesting.
