I am assuming you are talking about green anoles... There is only one certain way to sex Green Anoles and that is with a sexing probe. However there are a couple of physical characteristic that are just about 100% accurate. Mature males will have a hemipenile bulge at the base of the tail when viewed from the underside. That is where the hemipenes (sex organs) are located. Males will also have a pair of enlarged post anal scales that are clearly visible with slight magnification. If your animal has one (or both) of these characteristics, it is almost certainly a male.
The "white line" test and various others are not accurate since both males and females and juveniles may have white dorsal lines. Head shape, dewlaps and others are characteristic that may be shared by both sexes.

There are very good books that can help you, I have Anoles too, the book is called GREEN ANOLES BY JON COOTE. Heres what it says (theres pic.s too).
THE UNDERSIDE OF A MALE ANOLE SHOWS A PAIR OF ENLARGED SCALES ON THE BASE OF ITS TALE.
THE FEMALE DOES NOT HAVE ENLARGED SCALES.

As posted above, but also, in general, if you have two anoles and can compare them, broader head, larger dewlap, more 'masculine' appearance, it's a male. But without having two of the opposing sex, it's hard to tell if you don't get to see them all the time like I do. ;)

you should only be having sex with other people, yourself, or with human-designed toys! Leave everything else alone!
