Thanks everyone. I knew I needed another comma, I just didn't know the right placement for it. My handbook doesn't give any examples that reference this particular example closely enough.

Many would tell you to have it re-written with better grammar.

"We were drawn instantly to the puppy pens when Karen, the owner, opened the door to let us in."

I agree with Heather Y. artlogical's answer ends in a proposition ("in") which is grammatically incorrect.
