People's tastes vary widely. What I like and what I recommend might not suit your taste.
That being said, here are some of my favorite comedy/romances:
1) I've Heard the Mermaids Singing
2) Eternal Sunshine of the Spotless Mind
3) The Awful Truth (classic film w/ Irene Dunn and Cary Grant)
4) All About Eve (more of a drama, but really wonderful)
5) Central Station (Brazilian movie. Okay, it's not a comedy, but it's really really sweet)
6) The Straight Story (love story about 2 brothers. Very poignant)
7) June Bug

If you like movies with sad plots and tragic endings, try Titanic, Romeo+Juliet or Moulin Rouge. For comedy+romance, i would recommend 50 First Dates and Alot like Love. For romance with good endings, basically there are alot. But i like The Lake House most.
