16 hrs of straight driving, stopping only for petrol/toilet breaks (assuming 2-3 drivers) from Brisbane to Townsville, after that at this time of the year depends on the rainy season. Townsville to Cairns should be 4.5-5 hrs but could be up to 8 if the rains are bad.

One day with very few breaks.
It wouldn't take two, (like the others said) as we did the trip from a rural town in Vic, to Cairns and that took two (very few breaks).

brisbane to cairns is 1706.79km
drive wise that is 18hrs 47 mins on average

It is something over 2000km. You could do it in two days but it would be hard work and boring - and you would miss so much on the way.
