There are no direct train to Puri from Mumbai or Chennai..But you can get connecting trains.

If you want to Board from Mumbai,You have two options..Konark Express (daily) of Bhubaneswar-Kurla Express(weekly). If you use Konark Express then you can have journey till Khurdha-Road(this is a Junction for Puri),But I seriously advise to book ticket till Bhubaneswar because Khurdha-Road is big Junction but very small place and Konark Express reaches there at 3 .30 am early morning. Khurdha Road is just 15 mins away from Bhubaneswar.
From Bhubaneswar you have enough options!!You can use Bus(every 10 mins),Train(not less than 15 trains) or you can hier Taxi.
From Chennai, you can board any train connecting Chennai with Howrah and you can get down either in Bhubaneswar or Khurdha Road. Some of the trains are Howrah Chennai Superfast,Karamondol express,or Bhubaneswar-Chennai weekly etc.
There are enough places to stay in Puri from 4 star Luxury Hotels to Budget hotels. Also you can stay at Govt owned Pantanivas. There are also hundreds of Dharmsalas.
You need not stay at Konark as it is just 25 km from Puri and you can hier taxi or Bus to visit that place.
I will insist you to travel till Bhubaneswar as You can also visit Bhubaneswar and it is well connected with the two places you mentioned. Both Puri & Konark are just 60 kms from Bhubaneswar
