I don't know much about uni, even though I own it.

But basically, sims 2:

better graphics, more interactive.

I prefer it to the original any day

The Sims 2 has better AI and they take better care of themselves. The graphics are prettier. A major addition is that Sims have aspirations which you choose for them as you create them, the aspirations are things such as Riches, Romance, Science, etc. This is what they want out of life, and they have daily goals that raise their aspiration level which helps them get promoted in their careers. This is a great new function that adds some kind of goal to the game, but doesn't restrict freedom. Also Sims have family trees and they are more socially intelligent for example if a sim sees her friend's husband cheating on her then she will start stuff. It's generally deeper and sims have actual lives.

In the Sims university your sim has a new life stage : young adult. During young adult they can go to college and gain education as well as new friends. They live in a dorm and you have to get your sim through college socially and gradewise, you can affect it by talking to professors and building a relationship with them. It's something new.

The sims 2 has better graphics and you are directing a sims lifetime. Its more features also. And when you actually have a child you can see the genes from the parents. Its more stuff you can see if you go to www.sims.com

It's better and university is way better but it's an expansion so no biggie but there are extras.
