Lots of good answers here! For me, it's a toss-up. First, there's "The Age of Innocence" (w/Michelle Pfeiffer & Daniel Day-Lewis, Dir.by Martin Scorsese), due to the sumptuous production design, aching interpretation of the plot and the glimpse into 19th Century, American upper-class foibles.
The other is "Night of The Iguana", w/Richard Burton, Ava Gardner & Deborah Kerr, dir. by John Huston. It's (among other things) a scathing moral indictment of established religion, as study of mental illness and coping mechanisms, and how love can appear in the most unlikely situations...not to mention the triumph of a broken soul! It has everything, as most Tennessee Williams plays do!
Other faves: Amadeus, Dersu Uzala (by Kurosawa), JFK, Pleasantville, Ed Wood, Malice and the Ciderhouse Rules...to name only a few!
Whoops! How could I forget LOTR, esp "Fellowship"??!

Fear and Loathing in Las Vegas, anything by Stanley Kubrick, Saving Private Ryan, American Beauty (sorry to steal your answer, person above me), Man on Fire, Seven Samurai(or anything by Akira Kurosawa or with Toshiro Mifune in it)...
Jurassic Park(the original only, though I like the sequels), and various other films...
