I was given this target (from Edmund scientific), it looks like glass with Chrome markings on it (3 inch x 3 inch), and told to calibrate some microscopes. I have compound and dissecting scopes. Can I calibrate both with this target. And how the heck do I go about using this thing?

Not sure what they were referring to.

The USAF target is a RESOLUTION target, i.e., you can determine whether the microscope can resolve what it's supposed to, that's about it.

I've used such a target to verify the basic operation of video calipers, i.e., a length measurement system, but the USAF targets are not sufficiently well calibrated to actually calibrate video calipers, though.
