It depends on how you count it. The first American deaths at German hands occurred on 3 Sep. 1939, with the U-boat sinking of the British liner Athenia. The first overt act of war by the US on Germany was in April 1941, when the USS Niblack dropped depth charges on another U-boat, without effect. In June 1941 yet another U-boat sank the Robin Moor, an American freighter. On 16 October a destroyer group led by the Kearny engaged in an overt naval battle against a German wolfpack, and the Reuben James was sunk Halloween. All this was kept secret from the American public until December, when war was declared.
In the Pacific, you might consider the Panay incident 12 December 1937 as the beginning of hostilities, or the introduction of Chennault's AVG to the Chinese air battle. Or even the oil embargo in the summer of 1941 that precipitated the Japanese plans for the date that's most commonly used as the start date: 7 Dec. 1941, the Japanese attacks on Pearl Harbor and other Pacific sites.

While many Americans felt the need to leave the country and join foreign armies to help the British and Canadians fight the war. The US did not get involved in WW2 until the Japanese bombed Pearl Harbor. The fears from the first WW lingered heavily with the American people and it took Pearl Harbor for the people to be behind Roosevelt enough to allow him to fight back. Once he declared war on Japan, they included Germany because Hitler had manufactured a faux alliance with the Japanese government.

It was the day after Pearl Harbor which was Dec. 07 1941. On Dec. 08, 1941 President Roosevelt asked the Congress to declare war against Japan. After the declaration of war against Japan, Germany as an ally to Japan, step in and also declared war against the US. At the end of Germany's declaration, the Congress then declared war against them. Thus, the beginning of all out battle against Fascism and Nazism.
