I think the best way to answer this is to do it like a politician would. You want to use the question to show the improvement and progress you have made in the past. For example, when someone asks you what your weakest trait is you can say, "Well, in the past I have had a little trouble staying organized. But I learned that it really helps me to write everything down and go over it at the start of each day and prioritize everything." This shows that you can identify your own weakness take steps to improve upon it. It also shows you are a progressive and self-motivated person.
I would not suggest answering with blatant honesty because you don't want to make yourself sound bad if you can help it. Also, I would not recommend saying something about how you work too hard and that's your downfall. Interviewers hear this all the time. It's pretty much a cliched answer and sounds cheesy.

Before you go on a job interview, make a list of your strengths and weaknesses. When an interviewer asks you about your weakest trait, you will be able to tell them. Practice before you interview what you want to say, that way you can avoid any duhs and uhs. You will be able to give the interviewer a straightforward answer.

I speak what is on my mind, not thinking about the outcome.
If you ask for my opinon i will be flat out honest.
A really cheesy one that always works is, i am too decicated to work and that i always care about my employees and families needs before mine.

The best way is to definitely answer it and put a +ve spin on it. Something like - "I have been told by my friends and coworkers that I get way too involved with my work at times and neglect my personal life."
This will show that you are passionate about your work and give in your 100% when needed.

Pick something that is actually a good trait but say you're a real stickler about it. For instance:
"Well, I am an extremely (organized-prompt-neat-detail oriented- positive- etc.) person and sometimes I find myself getting impatient for people who aren't."
