In theory, you can. There are practical problems, such as the cost of constructing something that may never be used, and the cost of maintaining it. The extent that such a ship could sustain itself without support is still unproven, despite visions of plants growing in veggie bins and being able to generate oxygen and energy from wastes. There are also moral problems, such as who will select the people to go on the ship.

Hi. Possibly, but you need to decide how many folks would be required to prevent inbreeding if they want to repopulate the Earth.
