Absolutely. I am a marketing professor and I've written letters and gotten calls for everything from the Marine Officer Corps to elementary school teaching for former students. It does help if you didn't graduate too long ago and/or are still in touch with the professor. Just call or email them and say that you were applying for a job doing XXX, and you were wondering if you could use them as a reference. Usually for jobs, they just will be agreeing to take a phone call from the employer. Then give the professor your resume and any other information they may not have about you (like what you have been doing since taking their class), and explain to them why you are applying for this job.

Call the college. Ask for the professors extension and talk to him. Just tell him/her that you are applying for a job and need a Character reference. It would be counted as a character reference and not a former job reference.

Call or send an email. If you did not know them on a personal level, remind them of the classes you had with them or maybe send them a paper you wrote for one of the classes. If they need to mail the reference, provide them with a stamped and pre-addressed envelope. Professors get requests like this all the time, so just be polite and professional. Good luck with the job!

I think it is acceptable. It is preferable to have at least three references, and one might not be enough.
