Can a wireless access point, attached to a second computer, connect to a wireless router connected to the main computer?
Or, must I use an internal PCI card wireless device on the
second computer

Main computer was used to install the Wireless router ma be, but once the router gets installed you should be able to access it from that other computer as long as you have a wireless card or a Wireless USB Adapter installed on it.

I am assuming you have a working internet connection through your wireless router, and your main computer can access the internet fine. I am also assuming your second computer has no wireless ethernet card, but does have a wired ethernet card, and you don't want to run wire to your existing wireless router (or your wireless router is one of those evil models that has no wired ethernet LAN ports). I am guessing you want to connect the wireless access point to your second computer and connect wirelessly to the existing router.
This will not work unless you can get the wireless access point to function in client mode instead of the typical infrastructure/access point mode. For example, read the documentation for the Linksys WAP54G.

why not just connect both computers to the wireless router? you need a wireless card in both computers regardless to talk to the router or the access point.

Yep usually you just use a cross-over cable between the access point and the computers network card. You will need to look for the feature to do this, but it can be done.
