Yes it does.

To start off, a heavier cart Will take more energy from the pushers to get up to speed.

Secondly, depending on the bearings at the wheels, a heavier cart will have more friction on the road but the pull from the earth is the same for a heavier and lighter cart. If everything was frictionless the weight would have made no difference.
