I have recently purchased some speakers. [Durabrand 2.0 CH USB powered speaker system]
They are supposedly PnP however when I connect both the USB and the sound card cord into the computer it does not connect either. I have a usb flash drive that I have tried with the same USB port I am using and it does detect it, So I don't think that the usb port is messed up. Can someone help me, please?

You might be tring to use too much power. Some usb ports have full power (500ma), some don't.

Try a different usb port, or use an ac adapter with the speakers to see if that works.

(To check, right click My Compter, Device Manager, devices, click on the usb hub(s), Power tab)

You can add a powered usb hub if this is the case.

Also - try the speakers on another pc. They might be bad!

If the speakers plug into BOTH usb and the head phones jack, then it doesn't need to detect anything. It's getting the sound from the headphone (or speaker out) jack like a set of headphones and is using power from the usb jack instead of a wall outlet. If you can't get any sound, try plugging it into the headphones jack and turn the volume up on your pc.

I think you can't use two speakers at the same time ( Speaker that is connected into your soundcard and the one you just bought) you can use one speaker at one port at the same time.

Or if the speaker that is connected into your sound card is disabled and you want to use the USB speaker try to test it into someones computer if it is working perfectly
