already have software background but want to brush up on programming skills, which language would be best to take a course in and really develop my skills for......java, asp.net (with c#), php, or c++

That would kinda depend on what kind of employer / industry work you are seeking... Wouldn't it?

A lot of employers have been using Java in the past, but there is a large shift toward using ASP.NET (with C# or VB). This is because of the great controls for displaying data. It makes handling data a lot easier and allows them to get more production from less work. The learning curve for all of those languages is similar. If you learn Java, C#, VB - all of these are very similar in most aspects, especially Java and C#. C++ is considered outdated by many people. PHP is nice because it is free, but at the same time it is not as user friendly as ASP.NET.

Java or .net with sql experience is great. A lot of major companies are shifting toward web services and xml. Anything you can do to get experience in any emerging trends is great. In the last year or so AJAX has become a web programming buzz word for employers.

i think that employers mainly use java since java is a real portable programming language that has rich libraries thus supporting a wide number of scope. c++ is also good since it is native to machine and platforms and tend to be more machine efficient than java.
