I am not a selfish person by nature, so I've never felt comfortable with making wishes, even the ones you make after blowing out your birthday candles. If I want something better for my life, than it is my responsiblity to make it better. By ponding that responsiblity off to a genie to take care of, that is a the coward's way out and also what ever you wished for the value would be greatly diminished because you didn't attain yourself, but it was merely handed to you.
But if it's only one wish I get, than I would wish that what ever path I choose to walk upon in this life, that I am at peace with my decision and that through my own hard work I become successful and content (happiness is a goal few people are truly worthy of attaining - so I will gladly take contentness).

To know all that there is to know.
With this knowledge I could cure the world of all the diseases.
Knowing everything there is to know would also allow me to have all the money I wanted because I would always know the winning lotto numbers, horse races etc.
I would also know the outcome of every decision.

That my beautiful son will live longer than I do. It is my biggest wish, and often times, my biggest fear when I think about it not happening.
