Oh I love Prague - just got back from going again a few weeks ago.
If you've never been before, go to Old Town Square and just pick up one of the walking tours they do - best bet to see everything. Apart from that... Old Town Square is great place to start - check out the astronomical clock, head just a bit out of the square to the Jewish Quarter, see the Jewish Cemetery etc Then Charles Bridge, up the hill to the castle. Wenceleas Square is lovely with the National Museum. Hey and try the sex machine museum just off the square!!
Just walk man - you'll see everything and get the vibe. Don;t head too far out of town though.If you love Mucha or Salvador Dali, theres an exhibition of both in the Old Town Square at the moment. If you have enough time - get opera tickets!
Nice places to eat include Pravda and if you are a vegetarian, Lehka Hlava on Borsov 2 which is a little and very funky veggie restaurant. Bars you have your pick of but avoid the ones in the main tourist areas if you don't want lager louts!
Watch your wallet as pick pockets are rife and agree a price in a taxi before you get in - watch the ones around Wenceslas Square as they are a bit dodgy.
But if its your first time I'd recomend doing a tour so you don't miss anything.

Gwen has great ideas. I would also recommend checking out some of the walking tours. they meet at the main square every evening at about 7 or so (there's a website somewhere). the haunted Prague one is really fun and hey have a pub crawl on that is great!!
Depending on haw long you'll be there, I would also say that a day trip out of the city is really fun. Karlovy Vary is a great place for spas, and Kutna Hora is a half day trip to the "Bone Church", that's really made out of bones.
Depending on where you are staying, you may not need to worry about taxis, you can ride the tram or metro system. Both are easy and efficient.
Walk a few blocks in any direction and go into whatever pub looks cozy. All are a good time. Also, don't forget Mala Strana on the Castle side of the river. There's a great place for jazz called U Maleho Glena, and the Blue Light Bar on Josefov.
My favorite restaurant by far is called Maly Buddah. It's a cab ride from the center ( have the guy drop you at the top of Pohorelec) and walk down the street, the only direction) and it's about .25 miles on your left. Fantastic fresh asian food, in a dim lit, cave type atmosphere. nice wines and tea. After dinner you can walk across the street to this lookout point and get a view of Prague at night.

Try dry bob sleighing, a shooting range and if your in to football go to a Slavia Prague game its around £1.75 to get in and around 50p a beer in the stadium and the atmosphere is second to none.

Go up that big hill, there are pubs with pints of beer at 20p.
They close early on weekdays though, at about 10pm, but weekends they open late.
Also, just walk around the centre, it's really beautiful.
