I can't help much with Sydney, but here are some ideas for Singapore:

Shopping: Orchard Road
Evenings: Clarke Quay, Little India, Chinatown, 'bum boat' trip on river (start Clarke Quay)

Others:
Botanic gardens, Sentosa Island (take cable car one way at least), Johor Bahru for sea food, Zoo (but don't bother with bird park anymore), City Hall and Raffles Shopping Centres, Bugis Junction shopping.

History: a few sites like Changi Gaol and the 'Battlebox" on the hill above Clarke Quay

If you are in Sydney on 3rd March, don't miss the Sydney Gay and Lesbian Mardi Gras. The parade is spectacular and starts around 8pm. Unless you get a space by the barricade very early, avoid Oxford Street and lower Flinders St if you want to see anything.

How long are you there for, what kind of things do you enjoy and how much are you willing to spend?
There's no point me telling you about museums if you hate museums, and no point telling you about the surfing spots if you can't swim. If you give me some indication of what you enjoy i can tell you might find fun in Sydney - there's a lot to do there!
