To quote Wikipedia, LEGO MINDSTORMS is a "line of LEGO sets combining programmable bricks with electric motors, sensors, LEGO bricks, and LEGO Technic pieces (such as gears, axles, beams, and pneumatic parts) to build robots and other automated or interactive systems".
There are two available versions of MINDSTORMS; the nine year old RCX and the NXT from mid-2006. Among the advantages of the NXT over the RCS is better hardware on the programmable brick itself, Bluetooth support (imagine controlling the robot with your mobile phone), much better sensors and motors and a much more intuitive programming environment.
The equipment needed to play with MINDSTORMS is one of the two sets (I would recommend the NXT) and a computer.
The way it then works is that you build your robot out of LEGO bricks (including motors and sensors), use the computer to create a program for the robot (controlling input from the sensors and output to the motors), upload the program to the robot and then start the program on the robot.
You can then build anything, and even participate in various competitions. One such notable competition is FIRST LEGO League, a science competition for children where one of the tasks is to build and program a MINDSTORMS robot to perform certain tasks.
