Everyone said it best: call a cab. However, if you're intent of driving here then I've posted a link for a really good road map of St. Thomas. Basically, you get on route 30 and follow it all the way past Havensight. Eventually you'll reach a fork in the road with a stop sign. Instead of taking route 30, follow route 315 all the way to Marriott. Oh, and don't ask anyone here for directions because we'll only give you landmarks. :)

I'll add to Hector's answer by saying:

Take a right out of the airport access road.
Follow that road thru town.
At the big new Yacht Haven Grande Marina you bear right ( as opposed to taking a hard left).

Follow that road past the cruise ship dock.

When you get to that fork in the road Hector mentions....it's at the top of the hill you climb getting out of Havensight...the cruise ship dock area.......the stop sign isn't that clear. It will look like the road you are on goes straight ahead to Frenchmans Reef ( and it does).

The problem is its considered a turn; traffic coming from your left and turning in front of does NOT stop and will wallop you into next week if you don't stop and look left.

If you do this drive on a Thursday or busy cruise ship day, allow 45 minutes to an hour to go the 6 miles from the airport to the Reef.

Call the hotel. They will give you the ph# of the taxi service that they deal with. That way you can be picked up when your flight lands and be the only passenger. Otherwise, there will be many many many ( I cannot stress how man) taxi vans sitting out side of baggage claim when you arrive. There will be taxi assoication stands also located at baggage claim that can tell you exaclty what the flat fee would be to the hotel that you want to go to. It is better to bargain with the drivers directly for a better price. If you just dont want the hastle or to wait when you arrive or you want a private ride with no other guests/riders then you should go through your hotel.

The easiest way is to take a taxi. The taxi association has people right outside from where you get your luggage, and they will tell you which cab to get into. The cabs are more like "busses", they will be taking other people other places as well. And there is a baggage fee. but the rates are set and posted so you don't need to worry about haggling over the fare.
