There are two main ways. They either look at a large collection of facts and data and search for patterns in them that can lead to a hypothesis, or they become familiar with a body of scientific work, and use that foreknowledge to develop a hypothesis and test its validity. Normally hypothesis are developed through either direct observation, statistical observations, or calculated observation. If you notice on the scientific method, the first step is observation and writing those observations down. The second step is the formulation of a hypothesis. So without observation, scientists cannot develop a hypothesis.
If the scientist uses other peoples theories as a basis, he/she is also using their observations as a basis.

They study existing theories and the results of experiments, then try to think about the inconsistencies and possible problems. Often hypotheses come about as a result of trying to understand some anomolous result of an experiment, and then getting an idea in a flash of creativity. Then the hard work starts---the hypothesis needs to be scrutinized and subjected to test.

Hypothesis is a guess that scientists make about a certain subject. I know this because this year we did a science project (each individually) and it was preety easy for me. Maybe because I am in 6th grade.

They use all of their training and previously gained knowledge to make an educated guess as to what is happening. Occasionally, they will be totally stumped and will sometimes attribute clever or unique solutions to a kind of "divine inspiration".
