The party would have to choose someone else to act as Prime Minister. The voters only decide the party, not the leader... (sadly)

The current Government would not be reelected - a new Government is always formed after a General Election. You mean that Labour would win the most seats but the PM has lost his. In this case the Labour Deputy Leader would be the leader of the majority group in Parliament and would therefore be invited to form a Government by the Queen.

It is actually not necessary for a Prime Minister to be an MP. In Britain, the Prime Minister is chosen by the head of state, and the head of state is the Queen. Theoretically, the Queen may choose anyone she likes to be Prime Minister, whether or not he or she is in parliament. If she wants to choose you, she can.

But a Prime Minister has to be able to form a government, and he or she can only do this from among the MPs and Lords in Westminster. So, unless you could persuade enough of them to support you, you would have to go back to the Queen and tell her you had failed to form a government. She would then choose someone else.

Plenty of people without seats in the House of Commons have been Prime Minister - those that have been chosen from the House of Lords. But these days it would be very difficult to retain public support for an unelected Prime Minister. In practical terms, if Tony Blair lost his seat, he would have to offer the Queen his resignation as Prime Minister, and she would choose someone else from the winning party.
