The quickest I can think of is if you had an external modem to plug into it. Modems eat about any setting. You could bring up Hyperterminal on it and another pc and connect them with a null-modem cable and see if they talk. Or you could put an oscilloscope on pin 5 (ground) and look at pins 2&3 while you type characters in. (Don't really remember whether 2 or 3 is TD)

You cannot test it unless you have either a modem or other serial device to connect to, or a test (loopback) plug.
Either way, you open the programs->accessories->communications->Hyper terminal. There you define a connection name, then choose the serial port, then the connection parameters (speed,parity,stop bit,data control) and start typing. Depending on the device you should see some answer on screen to show the port is working.

Connect a known good serial device to the port and see if it will work with this machine. But be warey that if the device works on a machine with an older operatin system and not this one with a newer operating system the problem could be drivers or compatibility with the newer operating system.
