No, but if you put it in "standby" mode, it should still scan. (If it's off, it is as if your computer were in a coma, but if it's in standby mode, it's like it is sleeping lightly.)

In Mac OS, you can have the computer wake up, perform the scan, then shut down. Browse the control panels folder and you should be able to find everything you need to do that. I know there is, or was, a similar process for Windows, but I don't remember how.

No, the computer has to be on and actively accessing the hard drive to scan. So, it'd be better to either leave the computer on or change the scheduled time.
