It was Iftikhar Ali Khan who was made as a captain in his first match as an Indian. He was already playing for England before.

C.K. Nayudu was the first Indian cricketer to Captain Indian Cricket Team in the first test match played by him. His test debut was against England on 25th June, 1932. In the first Test Match played by India against England at Lord's from 25th to 28th June, 1932 Nayudu captained Indian Team.
