Mary, who HAS been in college for several years IS close to graduating.

Mary AND John who HAVE been in college for several years, ARE close to graduating.

(Although my sources say that John isn't really as close as he thinks!) :)

have; are

Mary and John are more than one person, so it's plural.

The words "have" and "are" follow appropriately.
