I would start with locally owned bookstores, not the big chains. Go in with your book and talk to the owner or manager. There's a local bookstore in our area that carries books from local authors. Quite a few of them are self published or published by lesser known local publishing companies.

Most self-publishing companies print on-demand, which means that the only way to get it on stores is if the owner of the store orders the book from the publisher. Most self-publishing companies give you your own author page and put your book on Amazon.com though.
If you can, arrange book signings and also talk to bookstore owners about your book. Expect to make very little money in self-publishing, it will probably cost you more than it will pay you, but at least your book will be published.

Generally, you can't.
Most self-published books are printed POD (print on demand). POD titles are non-returnable. And bookstores won't buy books they can't return.
You didn't say which service you are using. Most POD services have some sort of an ISBN service that will assign an ISBN to your title and get you listed in Books In Print and the Ingrams Database. If this is the case, generally most online booksellers will list your title as available. The bright side of POD is that since they require no inventory, most online vendors are willing to carry the titles on their sites.
If you are willing to invest in the books, you may have some success working out consignment deals with your area booksellers. You would eat all of the cost of printing the books, however, and would only get paid if they sell the copies.
If you are willing to make the investment, you may also try working out distribution deals with area book distributors. You would have to purchase copies of the book and send them to the distributor. Most have catalogs they provide to booksellers, who can then order your book. However, this is very pricey and very risky, as POD titles generally are expensive to produce to begin with, and you will need to be able to offer up to a 60% discount off the retail.

You will have to do your own publicity for the book. You can try to book yourself, on the local morning, afternoon and evening talk shows in your city.
Look online for how to write a Press Release and include it with a copy of the book, which you mail or hand deliver to your "local bookstores."
