Well, nobody can top Jimmy Hendrix. Ever. And his vocals arn't bad either. He's my favorite classic rock guitarist. I like Led Zepplin's guitarist too, but not as much.

favorite modern-ish guitarist is Ben Moody, formerly of Evanescence. Splitting was the worst thing that's happened to the band. They're still good, I suppose, but they're not the same since Ben left.

Django Reinhardt, an incredible French-club jazz guitarist, was probably the first guy who brought guitar to the forefront back in the 40s and early 50s, played with incredible feel for improvisation, impossibly beutiful and mellodic lines, and all that with basically two fingers only. No one can top THAT.
Of the more recent ones, Hendrix was great in terms of energy and innovation, but he wasn't much in terms of technique. Steve Vai and Joe Satriani are incredible, and the guy to keep an eye on is Jake Cinninger from Umphrey's McGee - in the last 25 years, he was the only one I heard that really made my jaw drop - and I have played for 26 years and seen Vai, Fripp, Belew, Clapton and many others live. He is amazing.

I have two that i can't pick between: Slash from gnr and John Frusciante from the peppers. Both are incredible and i also like david gilmour, brian may (amazing), and jack white is pretty cool too. the guitarists in my chemical romance have also proved their talent on their new cd the black parade, regardless of what anyone says about them. there's some wicked guitar bites from them on that album

Mike Stern, George Benson.. Jimmy Bruno.. Robben Ford.. Andre Bush.. So many.. I like "outside' type of playing.. Stern is good as is John Abercrombie.
