It's in the Republic of Mali (West Africa). Most Malians use the French spelling: Tombouctou or Tombouktou.
I almost went there once. I was working in Bamako, the capital of Mali, in 1963/64 and we decided to go on a long weekend trip to Timbuktu by Land-Rover. We got as far as Mopti (about two-thirds of the way) and realised that we wouldn't be able to make it and get back to Bamako in time to report for work on Monday. But we later heard that we hadn't missed much, as most of the historic buildings in Timbuktu had turned into piles of sand, largely through neglect. However, Mopti was a fascinating place to visit.
I've since heard that the Malian government has put a lot of effort into refurbishing the buildings and that Timbuktu is, once again, worth the visit.

Timbuktu (Archaic English: Timbuctoo; Koyra Chiini: Tumbutu; French: Tombouctou) is a city in Mali, West Africa. It is home to the prestigious Qur'anic Sankore University and other madrasas, and was an intellectual and spiritual capital and centre for the propagation of Islam throughout Africa in the 15th and 16th centuries.
Tombouctou or Timbuktu, city, central Mali, on the southern edge of the Sahara, just north of the great bend of the Niger River. It is connected with the Niger by canals and is served by the small river port of Kabara. The city is a regional trade center for salt and other basic commodities. Its few manufactures include cotton textiles, leather goods, and pottery.
