What are the differences in how they run and how they work?

The US Senate is elected. I don't know what they do.
The House of Lords is unelected. The British House of Commons is elected. They pass legislation, which then goes to the House of Lords for "sober second thought" review before the legislation gets Royal Assent (ie: the Queen signs it) and it's passed into law.
The House of Lords can delay the passage of legislation and return it to the Commons for further debate if they have problems with it, but it's very rare that they actually stop it from becoming law.
The House of Lords also does committee work and writes papers (I think they're called White Papers) on matters of national interest, making recommendations for how the government should act.

Members of the US Senate are elected (by the state legislatures up until 1917, but now by the people of the state), 2 per state. The House of Lords is hereditary, though some people think that the Senate should be hereditary, like the Kennedys and the idiots in Massachusetts who keep putting that drunken blimp back in Washington.

The house of lords is only open to a higher level and class of people.
The senate attracts a lot of scum.
The saying in the states, that the senate is perfect proof of the fact that every state has at least 2 idiots, isn't that far off. And it seems the bigger the state, the bigger the idiots they send. ;-)
Think California, Texas and NY.

The House of Lords was originally elected only by the wealthy. Thats why they are the House of LORDS. The house of Commons was for commoners representation.
The US Senate has 2 members from every state. They are elected by states, not social class.

If I am not wrong Senate is Elected and Lords are Hereditary.The Senators have to work hard to run the country where as Lords have whole Day to drink single Malts and Shoot the shiiiite.
