Originally that is exactly what they did. Nowadays chemists interested in finding drugs with specific features work with extractives of plants know to contain specific chemicals with those properties.

Illicit drugs probably yes. Mainstream drugs no. They do analysis on safety. If deemed safe then they will pilot the drug with humans and or animals.
