I need a protocol for the preparation of plant chromosomes using the squash method. do u have one?

I'd love to help you out, but I don't know one. On the other hand a GREAT protocol for viewing plant chromosomes is a nitrous oxide method developed by Akio Kato in Jim Birchler's lab from the University of Missouri. check out Akio Kato's papers from pubmed for the protocol. I went from viewing 50 slides for a good spread to viewing just one and getting lots of good cells. JC Lamb also has a lot of papers out using that same method.
